Castelletto rose in the medieval period as a fortified site, to protect the border of the Aleramico Marquisate. In the 12th century it rebelled against Barbarossa and joined the Lega Lombarda. In the 13th century it was contested by Alessandria and in 1290 the militia of Alessandria fought against those of the Marchesate. The town was devastated in 1515 by the Spanish army and in 1642 by the Lanzichenecchi. In 1691, it passed to the Marquises Beretta Landi, originally from Milan and later to the tenant Antonio Maria Miglietti of San Salvatore. Castelletto becomes a municipal community towards the first half of the 18th century, a time in which the parish Church of San Siro was built at the top of the village.
The primitive feudal castle, centre of the current village, with the denomination of Castrum Scatiosorum, namely Castelletto Scazzoso, is mentioned as the property of the Scazzoso/Foresto family, originally from Vercelli. In 1937, the name of the village was changed by a Royal Decree to Castelletto Monferrato.
In the 20th century, Castelletto became a significant example of residential promotion of its hilly and highly attractive landscape, among vineyards and cultivated areas. This is demonstrated by the birth and development of "Villaggio Valverde".
The typical products of the rich surrounding countryside include the pumpkin, used to make delicious lasagne pumpkin and gorgonzola.
